- Please consider this as one of the parameters when designing primers.
- Tm is the temperature at which 50% of the double-stranded DNA dissociates into single-stranded DNA.
- It is said that the reaction works best when the annealing temperature for PCR is set at a temperature about 5℃ lower than the Tm
- The annealing temperature for PCR is typically set between 55℃ to 60℃.
- The values of ΔH and ΔS are taken from the reference.
- By checking the box for “Multiple Sequences“, you can calculate the Tm of multiple primer sequences at once.
